I came across this information on the website - http://www.saranagati.net/index.php?S=3&Article=269

Thought it might be of some help regarding your question -

Chanting: Which Side to Start From?

Chanting should begin from the big bead of your japa mala. That side should have a thread after eight beads to signify the eight principal gopis (asta sakhi). H.H. Jayapataka Swami was present when Srila Prabhupada refused to give initiation beads to someone who did not have the string after the eighth bead. He said they were not bonafide without the string. When asked why, Srila Prabhupada replied, "The beads represent 108 gopis and the eight signify the eight principal gopis. Chanting starts from the large size."

If you do not have a thread on your mala, it is not too late to braid one on now.

Also, I have been made aware by Rasa Prabhu in the Japa room that Srila Prabhupada first instructed the devotees to chant 64 rounds which were then gradually reduced to 16 rounds.

You can visit the japa room every saturday at www.krishna.com where Rasa Prabhu answers all questions regarding Japa
